Selection of DNA markers for detection of extreme resistance to potato virus Y in tetraploid potato (Solanum tuberosum L.) F1 progenies

Abstract: Abstract:Marker-assisted selection is progressively introduced into potato breeding especially in the case of monogenically inherited traits. Varieties and clones with extreme resistance to potato virus Y (PVY) from German, Hungarian, Polish, Dutch and Slovak breeding programmes were used as female parents in 6 crosses. F 1 progenies were subjected to the bulk segregant analysis. Polymerase chain reaction (PCR) based markers for Ry sto and Ry adg viral resistance alleles were used for the evaluation of varieti… Show more

“…Consequently, the 13 cultivars that were found to be resistant to both strains of PVY are likely to possess one of the Ry extreme resistance genes. It was likely that at least Ry sto was present, as it has been previously found in the cultivars White Lady and Rioja [15,25,38], which were both found to be phenotypically resistant here.…”

“…The phenotypic scores for cultivars from this study are generally consistent with the results reported from other studies. The cultivar White Lady was also reported to be resistant to PVY in four previous studies [15,25,32,38], and Cernák et al [25] also reported Rioja to be resistant. A number of cultivars, including Atlantic [31,39,40], Bliss [37], King Edward [31,39], Kipfler [37], Pike [31], Ranger Russet [19], Russet Burbank [19,31], Shepody [8] and Umatilla [19], were reported to be susceptible elsewhere.…”

“…Similar findings were reported by Flis et al (2005), who were unable to detect any of their markers in this variety. Heldák et al (2007) were also unable to detect the markers reported by Flis in Santé. These authors also attributed their findings to the causes outlined above.…”

“…Thus, for more efficient use of the Ry gene in potato breeding, the knowledge on the allelic dosage of this gene in the parents available in the breeding program is crucial (Koeyer et al 2010). One way of estimating the allelic dosage of a gene in a particular genotype is by the analysis of the segregation of this gene in the progeny, obtained by test crosses, i.e., with a nuliplex parent (susceptible) (Ribeiro et al 2006, Heldák et al 2007, Ortega and Lopez-Vizcon 2012, John et al 2013). …”
