Selective Loss of Natural Killer T Cells by Apoptosis following Infection with Lymphocytic Choriomeningitis Virus

Abstract: Natural killer T (NKT) cells, a unique subpopulation of T cells, coexpress markers also present on NK cellsand recognize the major histocompatibility complex class I-like CD1d1 molecule. We studied the effect of an acute virus infection on NKT cells. Mice were infected with the nonhepatotropic Armstrong strain of lymphocytic choriomeningitis virus (LCMV), and at various times postinfection, mononuclear cells from the liver, peritoneum, and spleen were isolated. It was found that within 2 to 3 days, there was a… Show more

“…In brief, lipids resuspended in PBS (pH 7.4) were mixed with the dimers and incubated at 37 C overnight. Mouse liver mononuclear cells, isolated as described previously [48], were treated with 2.4G2 hybridoma supernatant before staining for cytofluorography with empty or lipid-loaded CD1d dimers, FITC-conjugated anti-mouse-TCRb and biotinylated anti-NK1.1 (Pharmingen). The dimers were directly labeled with phycoerythrin using a mouse IgG 1 -specific Zenon labeling kit following the instructions of the manufacturer (Molecular Probes, Eugene, OR, USA).…”

“…Splenocyte, liver MNC, and PEC isolation was described previously [28]. Peripheral blood was collected from euthanized mice via cardiac puncture and treated with 0.5 mM EDTA and stained with the appropriate mAb.…”

“…The role for NKT cells in the pathogenesis of infectious diseases is less clear. However, the CD4 + V § 24 NKT cells were recently found to be extremely sensitive to HIV infection [11,23,24], and the corresponding murine V § 14 NKT cells are lost during acute infection with lymphocytic choriomeningitis virus (LCMV) [25].…”
