Sensitive and selective amplified visual detection of cytokines based on exonuclease III-aided target recycling

Abstract: A new Exo III assisted strand-cleavage cycle and ligand-responsive quadruplex formation strategy for amplified and label-free detection of IFN-γ was reported with a detection limit of 0.1 pM and a visual detection limit of 20 pM by the naked eye.

“…Recently lots of research work on hairpin DNA probe based immunosensors for the ultrasensitive detection of biomarkers has been reported. (Ge et al 2016;Gong et al 2014;Guo et al 2015;Yao et al 2014;Zhang et al 2014a;Zhang et al 2014b) This topic has been symmetrically reviewed. (Huang et al 2015b) As an example, a hairpin aptamer DNAzyme probe was used for sensitive and visual detection of IFN-γ based on an original quadratic amplification strategy ( Figure 5).…”

“…The peak at $430 nm was corresponded to the absorption of Ag nanoparticles, only the peak at $560 nm was the characteristic absorption of the DNA-Ag NCs. 3 So we chose 560 nm as the wavelength of excitation to measure the emission spectra of the TBA2-Ag NCs (Fig. 1C).…”

“…During the past decades, DNA-templated silver nanoclusters (DNA-Ag NCs), 1 as a new class of promising uorescence nanomaterials, have been extensively used in biosensing [2][3][4][5] and bioimaging. 6,7 Compared with traditional luminescent probe, 8,9 DNA-Ag NCs show advantages of stability, biocompatibility, ultra-small size, lower toxicity, and a strong uorescence emission.…”

“…Such molecular beacons-assisted amplification method, with its inherent stability, specificity, and simplicity, has recently emerged as a potential amplification technique for rapid and cost-effective detection of protein. On the other hands, several promising isothermal signal amplification strategies also have been introduced for the protein assay (Hao et al, 2014;Zhang et al, 2014), such as telomeric repeat amplification protocol (TRAP) (Liu et al, 2015) and nicking enzyme signaling amplification (NESA). Recently, several methods devised utilizing the advantages of molecular beacon and the isothermal exponential amplification reaction were proposed (Duan et al, 2013).…”
