Separation of triacylglycerols in a complex lipidic matrix by using comprehensive two‐dimensional liquid chromatography coupled with atmospheric pressure chemical ionization mass spectrometric detection

Abstract: The present investigation describes the employment of a comprehensive 2-D HPLC system, based on the combination of a silver ion and an RP column, for the characterization of the triacylglycerol (TAG) fraction of a very complex lipidic sample: donkey milk fat. The TAGs were grouped on the resulting bidimensional contour plot according to their double bond numbers (aligned along vertical bands) and according to their partition numbers (aligned along horizontal bands). Peak assignment was supported by using atmos… Show more

“…In our study, the employment of a partially porous column at a flow rate of 4 mL/min allowed to reduce the modulation time from 120 to 90 s in comparison with the previous reports [13][14][15]. The separation obtained for the TAGs fraction of a borage oil sample on a short partially porous column is shown in Fig.…”

“…The attempts concerning the on-line connection of reversed phase (RP) and silver ions (Ag) columns, denoted as 2D LC, [24][25][26] and supposed to technically simplify the off-line analysis, 27 found limited application. At present, RP-HPLC and Ag-HPLC columns combined off-line and connected to a mass detector appear to be the most powerful tool in the analysis of complex TAG samples, as demonstrated by Holcapek and coworkers.…”

“…Monolithic columns are ideally suited for implementation in the second dimension due to their high permeability and short regeneration characteristics [73,74] and have therefore been widely exploited in comprehensive LC [47,[75][76][77][78][79][80][81][82][83][84][85][86][87]. Furthermore, they allow operation at elevated flow rates without loss in resolution.…”

“…The loop interface is the most widely used interface and is based on a two-position/10- [47,48,71,[77][78][79][80][81][82][83][84][85][86][87][88]102,[106][107][108] or 8-port [57,83,95,96,[109][110][111] switching valve equipped with two storage loops with identical volumes. Other configurations have been developed using the same principle through a combination of several 6-port valves [61,84,85,112].…”
