Serological and Molecular Investigation of Brucellosis in Breeding Equids in Pakistani Punjab

Abstract: Brucellosis is an important zoonosis worldwide. Equines are susceptible to the infection when in close contact with infected animals. The objective of our study was to update the existing knowledge and detect and differentiate the causative agent of brucellosis in breeding equines in Punjab, Pakistan. A cross-sectional study was designed to evaluate the occurrence and etiology of the infection in the equine population in three districts. A total of 448 equine sera were collected from three prefectures viz. Sah… Show more

“…Thus, the diagnosis of brucellosis has to be accompanied by molecular detection and/or culture in endemic countries in humans and animals. Although brucellosis has been extensively investigated in farm animals in Punjab [23][24][25][26], this disease in humans is not well investigated yet. Few studies were carried out relying on serology and PCR, while studies using/applying culture and subsequent identification are next to none.…”

“…Previously, 21.4% prevalence by RBPT of brucellosis and 3.56% by ELISA in equine animals kept in different districts of Punjab province, Pakistan, have been recorded [ 23 ]. In our study, no significant difference was recorded in the prevalence of disease based on different age groups of cattle.…”

“…We drew 5 mL of blood from the jugular vein of all the study animals separately. According to previous procedures, all the serum samples were subjected to RBPT and ELISA to detect brucellosis [ 23 , 47 ]. We used an automated hematology analyzer for hematology.…”

“…So far, B. melitensis has been confirmed only by real-time PCR in cattle and buffaloes but not by isolation (62). In addition, B. abortus has been diagnosed only by real-time PCR in sheep and goats, horses, camels, and dogs without confirmation by isolates (32,36,69,72). In addition, in wildlife, only anti-smooth-lipopolysaccharide (LPS)-Brucella (B. abortus and B. melitensis) antibodies have been reported without any PCR-based detection (73).…”
