Serological characterization of small ruminant lentiviruses: A complete tool for serotyping lentivirus infection in goat

Abstract: Small ruminant lentiviruses (SRLVs) form a very heterogeneous group of ssRNA viruses able to infect goats and sheep worldwide. The genetic heterogeneity is reflected by a large antigenic variability which can represents a potential bias in serological diagnostics. Indeed the circulation of four different viral genotypes reveals how the surveillance and control of SRLV is a hard challenge. In previous works we described the use of a single subunit of the viral capsid protein for the SRLV infection genotyping. T… Show more

“…Infection by divergent SRLV strains in these animals is unlikely since primers used in PCR were designed based on known genotypes. Instead, low or fluctuating antibody titers may account for this discrepancy between serological and molecular techniques (36,37). Indeed, absence of serological response has been described in the periparturient period (38) as well as a result of recent infections (39).…”

“…After a final washing step, the reaction mixture was developed with 2,2′-azino-bis-(3-ethylbenzthiazoline-6-sulfonicacid). The results were read at 405 nm and the absorbances were obtained for each serum sample against the screening antigens and the three subunits [18].…”

“…The investigation of genetic heterogeneity in these epitopes among the different SRLV strains could reveal diagnostic drawbacks of the existent serological diagnostic tools and contribute to the development of highly efficient ones [ 56 , 57 ]. Also, serotyping diagnostic tools discriminating the different SRLV genotypes have been developed using subunits of the viral capsid protein [ 58 , 59 ]. These tests could be used for the implementation of epidemiological studies in countries where the genetic characterization of the circulating SRLV strains has not been performed.…”
