Seroprevalence of Echinococcus granulosus infection in a Uruguayan rural human population

Abstract: Serological tests using hydatid cyst fluid (HCF) are useful as a primary screen for detecting asymptomatic infections due to Echinococcus granulosus in human populations. An enzyme-linked immunosorbent assay (ELISA) performed with whole sheep HCF was used to determine the prevalence of hydatid antibodies in a rural human population in Uruguay. In order to eliminate cross-reactions with sheep proteins in HCF, 1% normal sheep serum was added to each human serum before testing. ELISA seropositives were further te… Show more

“…The mean follow-up period was 4.4 years (range 3 to 5.8 years). Immunodiagnosis Anti-echinococcus antibodies were assayed in human sera using a peroxidase micro-ELISA (enzyme-linked immunosorbent assay) [4] and modified by us [2]. Circulating antigen (CAg) and immune complexes (CIC) were assayed in whole human sera using a capture antibody method [5].…”

“…Hydatid disease, or human cystic echinococcosis, is caused by Echinococcus granulosus and is a public health problem in Uruguay [12,15]; some reports indicate a prevalence higher than 1% in the human rural population [2,13,14].…”

“…Serologic tests for antibodies to Echinococcus in human sera. Serum samples from those individuals who demonstrated a cyst on ultrasound were tested for total immunoglobulin antibodies by micro-ELISA using crude hydatid cyst fluid antigens as previously described, 14 and for total IgG, IgG1, and IgG4 antibodies using purified antigen B from hydatid cyst fluid. 30,31 In addition, a Western blot test using crude sheep hydatid cyst fluid was used to check for specific antibody (IgG) binding to the low molecular weight (between 8 and 20 kD) hydatid cyst fluid antigen B subunits.…”

“…13 Similarly, seroprevalence studies in rural communities, using an ELISA with crude sheep hydatid cyst fluid antigen to detect antibodies (total immunoglobulin) revealed an overall seropositive rate of 1.2% (17 of 420). 14 Ovine hydatidosis has been reported in all of the 19 Departments of Uruguay with an overall prevalence rate of 34% at slaughter; a prevalence rate of 10.7% was also recorded in 5,500 dogs subjected to arecoline purgation (Orlando D, unpublished data).…”

“…To control the parasite, treatment of dogs with anthelmintic is the main procedure [8]. Some progress was achieved with this procedure and the prevalence in sheep was reduced significantly in some countries or areas.…”
