Serotonergic vulnerability and depression: assumptions, experimental evidence and implications

Abstract: In recent years, the term serotonergic vulnerability (SV) has been used in scientific literature, but so far it has not been explicitly defined. This review article attempts to elucidate the SV concept. SV can be defined as increased sensitivity to natural or experimental alterations of the serotonergic (5-HTergic) system. Several factors that may disrupt the 5-HTergic system and hence contribute to SV are discussed, including genetic factors, female gender, personality characteristics, several types of stress… Show more

“…Indeed, serotonin (5-HT) transmission is implicated both in the onset of depression and in the mechanism of action of several antidepressant drugs (Jans et al, 2007). Moreover, findings from genetical and pharmacological studies indicate that serotonin signaling during early life is critically involved in the development of brain circuits that modulate adult emotional behavior (Ansorge et al, 2008;Ansorge et al, 2004).…”

“…Serotonergic vulnerability means that minor changes in serotonergic functioning do not cause symptoms but make the system more vulnerable so that additional challenges of the serotonergic system may result in the occurrence of psychiatric symptoms (Jans et al 2007b). Thus, serotonergic vulnerability can be demonstrated by challenging the 5-HT system, as is done with ATD; only vulnerable subjects will react to such manipulations with changes in behavior.…”

“…Serotonergic vulnerability (Jans et al 2006) is an increased sensitivity to natural or experimental alterations of the serotonergic system, such as genetic factors, female gender, personality characteristics (neuroticism), environmental stress (prenatal stress, life experiences) or drug use. An insertion/deletion polymorphism in the serotonin transporter-linked promoter region 5-HTTLPR modulates serotonin transporter gene (SLC6A4) expression, where the short allele (S) is associated with lower expression levels compared to the long allele (L).…”
