Serum antibodies to klebsiella capsular polysaccharides in ankylosing spondylitis

Abstract: Objective. To measure antibodies to Klebsiella capsular polysaccharides in the sera of HLA-B27 positive patients with ankylosing spondylitis (AS), compared with HLA-B27 positive and HLA-B27 negative healthy control subjects.Methods. Antibodies were detected by means of an enzyme-linked immunosorbent assay specific for each of the 77 known Klebsiella serotypes.Results. Significantly elevated frequencies and titers of antibodies to capsular polysaccharides K26, K36, and K50 were detected in sera from AS patients… Show more

“…However, if the organism is grown on the same media supplemented with pullulan (0.3% w/v) as the carbon source the degree of antibody binding in AS patients is greatly increased. This increase also shows that the capsule is highly antigenic and is one of the predominant antigens recognised by AS patients, an observation also made by other workers [14]. In the control experiment, where P mirabilis is grown in the same conditions, with and without pullulan, antibody elevations are only seen in RA patients not AS or healthy controls.…”

“…There is strong evidence for the involvement of humoral and cell-mediated immunity in the pathogenesis of AS, though investigations on abnormalities in immune cell numbers in peripheral blood [15][16][17], cytokine production [7,8,18] and occurrence of certain antibodies to different microbes [2,19,20] show conflicting results. It should be considered that there is a wide diversity of presenting symptoms in AS, and clinical expression varies a lot [21].…”

“…The parent K2 and K21a encapsulated strains were previously described (26). Spontaneous noncapsulated variants of serotypes K2, K21a, K26, K50, and K36 were obtained from nonmucoid segments of colonies of capsulated parent strains as described elsewhere (22,37). Likewise, a revertant capsulated K21a variant was obtained from mucoid segments in colonies of the noncapsulated K21a strain.…”
