Siansivirga zeaxanthinifaciens gen. nov., sp. nov., a novel zeaxanthin-producing member of the family Flavobacteriaceae isolated from coastal seawater of Taiwan

Abstract: A strictly aerobic, Gram-negative, rod-shaped bacterium (strain CC-SAMT-1(T)) showing gliding motility was isolated from coastal seawater of China Sea, Taiwan. Strain CC-SAMT-1(T) synthesizes all-trans-zeaxanthin (6.5 ± 0.5 mg g(-1) dry biomass) as a predominant xanthophyll carotenoid. As determined by 16S rRNA gene analysis, strain CC-SAMT-1(T) shared very high sequence similarity to the members of the genera Mariniflexile (96.1-95.3%) and Gaetbulibacter (96.0-95.9%); however, it formed a distinct phyletic li… Show more

“…Carotenoids were extracted and analysed according to Hameed et al (2012) with the following modifications: authentic all-trans-astaxanthin was used as a standard for qualitative and quantitative determination of astaxanthin; detection wavelength was set to 476 nm during reversedphase HPLC. Respiratory quinones of strains CC-AMO-30B…”

“…The number of representatives of the family Flavobacteriaceae is constantly increasing through the continuous description of new taxa. We have characterized species such as Lutaonella thermophila (Arun et al , 2009a), Muricauda lutaonensis (Arun et al , 2009b), Siansivirga zeaxanthinifaciens (Hameed et al , 2012), Kordia aquimaris (Hameed et al , 2013), Gramella planctonica (Shahina et al , 2014), Aquibacter zeaxanthinifaciens (Hameed et al , 2014a), Robertkochia marina (Hameed et al , 2014b) and Gramella oceani (Hameed et al , 2014c) from various marine locations in Taiwan. Here, we report on the taxonomic characterization of a novel zeaxanthin-producing flavobacterial strain, designated CC-PY-50 T , which was isolated from Formosan estuarine water.…”

“…The presence of flexirubin-type pigments was investigated as described by Reichenbach (1992) and Bernardet et al (2002). Carotenoids were extracted and analysed according to earlier descriptions (Hameed et al , 2012). The presence of the proteorhodopsin gene was tested using three pairs of degenerate primers (PR-Flavo-F and R, PR-Flavo-2F and 2R, and SA4-10 PRno3F and 3R) according to the descriptions of Yoshizawa et al (2012).…”

Susceptibility to antibiotics was investigated on MA using the disc diffusion method as described previously [38], and according to procedures outlined by the Clinical and Laboratory Standards Institute [39].…”
