Similarities and Differences in the Expression of Drug-Metabolizing Enzymes between Human Hepatic Cell Lines and Primary Human Hepatocytes

Abstract: ABSTRACT:In addition to primary human hepatocytes, hepatoma cell lines, and transfected nonhepatoma, hepatic cell lines have been used for pharmacological and toxicological studies. However, a systematic evaluation and a general report of the gene expression spectra of drug-metabolizing enzymes and transporters (DMETs) in these in vitro systems are not currently available. To fill this information gap and to provide references for future studies, we systematically characterized the basal gene expression profil… Show more

“…The relative expression levels of UGT2B7, UGT2B4, and UGT2B10 were examined in the HuH-7 and Hep3B cell lines. HuH-7 and Hep3B cells were chosen because both cell lines express multiple UGT2B enzymes (Guo et al, 2011) and endogenously express miR-216b-5p (Fig. 2), and Fig.…”

“…Furthermore, expression and functionality were confirmed for major P450s as well as for UGT and SULT enzymes, revealing donor-specific expression and activity levels comparable to plated PHH. In contrast, human hepatic cell lines, for example, HepG2 or HuH7, lack relevant hepatocyte functions (Andersson et al, 2012) and often show strong divergence in DME expression compared with PHH (Guo et al, 2011). The consistently high CYP2B6 Statistical analysis of CL H predictions from three different donors of sandwich-cultured UHH (151-03, 653-03, and 10-03).…”

“…The HepG2 cell line has been most widely used as a human hepatic cell model for toxicity studies. However, HepG2 cells are known to show extremely low activity of numerous xenobiotic metabolizing enzymes, [4][5][6] which could provide misleading results in toxicity tests of compounds that require biotransformation.…”
