Simultaneous determination of 10 kinds of biogenic amines in rat plasma using high‐performance liquid chromatography coupled with fluorescence detection

Abstract: We describe a simple, rapid, selective and sensitive HPLC method coupled with fluorescence detection for simultaneous determination of 10 kinds of biogenic amines (BAs: tryptamine, 2-phenethylamine, putrescine, cadaverine, histamine, 5-hydroxytryptamine, tyramine, spermidine, dopamine and spermine). BAs and IS were derivated with dansyl chloride. Fluorescence detection (λ /λ = 340/510 nm) was used. A satisfactory result for method validation was obtained. The assay was shown to be linear over the ranges 0.005-… Show more

“…Following fluorescence detection, a novel method allowing diagnosis of histamine intolerance by histamine and methylhistamine detection in urine by ultrahigh performance liquid chromatography coupled with fluorimetric detector (UHPLC-FL) was developed in 2017 . An HPLC method from 2018 with fluorescence detection allowed the determination of histamine together with nine other biogenic amines after derivatization with dansyl chloride with linearity in the range 0.25–10 μg/mL and a detection limit of 75 ng/mL for histamine in rat plasma . Coupling HPLC or GC with mass detection allows a decrease in the detection limit of histamine even more.…”

“…Pharmaceutical molecules containing nitrogen based functional groups like primary and secondary amines, imidazoles and also other groups like phenols were derivatized effectively with dansyl chloride and quantified accurately in biological and pharmaceutical preparations. [22][23][24][25][26][27][28]. The purpose of the current study is to demonstrate the efficiency of DNS-Cl as an suitable fluorescent labelling reagent for the new, selective and sensitive determination of TCG in pharmaceutical preparation and spiked human plasma.…”
