Simultaneous enantioselective determination of omeprazole, rabeprazole, lansoprazole, and pantoprazole enantiomers in human plasma by chiral liquid chromatography–tandem mass spectrometry

Abstract: Proton pump inhibitors, including omeprazole, rabeprazole, lansoprazole, and pantoprazole, achieved simultaneous enantioselective determination in the human plasma by chiral liquid chromatography–tandem mass spectrometry. The four corresponding stable isotope‐labeled proton pump inhibitors were adopted as the internal standards. Each enantiomer and the internal standards were extracted with acetonitrile containing 0.1% ammonia, then separated with a Chiralpak IC column (5 µm, 4.6 mm × 150 mm) within 10 min. Th… Show more

“…Among the cellulose‐based CSPs, cellulose tris(3,5‐dichlorophenylcarbamate) has the most applications in the chiral analysis of PPIs [75,82,90,93,123]. As commercially available immobilized CSP, it was used under both NP and RP conditions.…”

“…The validated method was applied to study protein binding ratio (except for ILA) using an equilibrium dialysis method. LAN, PAN, and RAB showed stereoselective plasma protein binding, while OME not [93].…”

“…Another aspect of the PPIs analysis is the chiral separation of a particular enantiomer. This issue has already been briefly described in the literature [ 67 , 68 ]. One example is the application of liquid chromatography and mass spectrometry (LC-MS/MS) for the simultaneous determination of omeprazole, pantoprazole, lansoprazole, rabeprazole, and ilaprazole enantiomers.…”

“…The developed method was also suitable in combination with equilibrium dialysis for the studies of PPIs enantiomers ratio binding with plasma proteins. The linearity of the method was determined in the range of 1.25 to 2500 ng/mL for the investigated enantiomers [ 68 ].…”

“…A thorough literature search dictates that some publications are available for the enantio‐separation of both the reported drugs [24–26]. But no paper is available on the high‐powered columns Chiralpak IA and Chiralpak IE columns using SFC and HPLC techniques.…”
