Simultaneous HPLC of twelve monoamines and metabolites shows neuroblastoma cell line releases HVA and HIAA

Abstract: Neuroblastoma is a solid tumor occurring usually in children less than 5 years old. It has been difficult to distinguish neuroblastoma from other childhood tumors through morphological diagnosis. Urine homovanillic acid (HVA), which is a metabolite of dopamine, has been proposed as a diagnostic index. Although increased levels of a serotonin metabolite, 5-hydroxyindole-3-acetic acid (HIAA), have also been observed in urine samples of the patients, they were largely attributed to dietary amines. By using an HPL… Show more

“…This may be the cause for severe hypertension associated with neuroblastoma [204][205][206]. Elevations in the levels of urinary metabolites of catecholamines vanillylmandelic acid and homovanillic acid are routinely assessed as prognostic biomarkers of neuroblastoma [207,208]. A report from the children oncology group suggested a correlation between the elevated expression of the vesicular monoamine transporter with clinical features, tumor biology, and metaiodobenzylguanidine (MIBG) avidity in neuroblastoma [84].…”

“…Ascorbic acid is used as standard. Antioxidant potential of the plant sample was quanti ied by executing the following free radical scavenging assays; DPPH, superoxide, nitric oxide, hydrogen peroxide, hydroxyl, reducing power and ferric reducing antioxidant potential (FRAP) by means of standard cited protocols (Blois, 1958;Liu et al, 2000;Klein et al, 1981;Ruch et al, 1989;Garrat, 1964;Oyaizu, 1986;Benzie and Strain, 1996). Percentage of inhibition or radical scavenging is directly proportional to antioxidant ability of respective extract.…”

“…Tissue samples were prepared and analyzed for biogenic amines and their metabolites, using reverse phase high performance liquid chromatography system (HPLC), with an electrochemical detector, as previously described (Cheng and Kuo, 1995;Liu et al, 2000;Chi et al, 1999;Auger et al, 2000). The apparatus used consisted of Waters pumps (Waters 600E Multisolvent Delivery System), 600 Controller, 717 Plus Autosampler and a 464 Pulsed Electrochemical detector (Waters, Milford, MA), and was controlled by a Dell computer using Mellenium 32 software.…”
