Single cell atlas of domestic pig brain illuminates the conservation and divergence of cell types at spatial and species levels

Abstract: Domestic pig (Sus scrofa domesticus) has drawn much attention from researchers worldwide due to its implications in evolutionary biology, regenerative medicine and agriculture. The brain atlas of Homo sapiens (primate), Mus musculus (rodent), Danio rerio (fish) and Drosophila melanogaster (insect) have been constructed at single cell resolution, however, the cellular compositions of pig brain remain largely unexplored. In this study, we investigated the single-cell transcriptomic profiles of five distinct regi… Show more

“…The collected tissues were rinsed by 1X PBS, then quick-frozen and stored in liquid nitrogen. The single cell nucleus of each tissue was separated by mechanical extraction method26 . Briefly, the tissues were first thawed, infiltrated by 1X homogenization buffer (containing 30mM CaCl 2 , 18mM Mg(Ac) 2 , 60mM Tris-HCl (pH 7.8), 320mM sucrose, 0.1% NP-40, 0.1mM EDTA and 0.2U/µl RNase inhibitor), then cut into smaller pieces, and the single nucleus was isolated by 2ml Dounce homogenizer set.…”

“…In recent years, the single-cell RNA-Seq studies of neurons isolated from various CNS components show distinct separations along the transcriptomic landscapes with individual clusters specifically linked to excitatory or inhibitory processes (Chen et al, 2019;Darmanis et al, 2015;Hu et al, 2017;Kupari et al, 2019;Li et al, 2020). Given the complex neural networks to and from the CNS that influence the ICNS, we compared our RNA-Seq data with RNA-Seq data from single CNS and PNS neurons to compare cell types across central and peripheral neurons.…”

“…There was also a small number of genes (n = 9) that were downregulated early, but were even further diminished at the two-week time point including Adra2a, Ccdc33, Dpp10, Fdft1, Hmcn1, Klhl14, Slc38a4, Tacr3, and Tfrc. Each of these genes have been reported as downregulated in models of chronic neurodegenerative disease such as Alzheimer's and Parkinson's 41,42,51,52,[43][44][45][46][47][48][49][50] .…”
