Single-Cell RNA Sequencing ofSox17-Expressing Lineages Reveals Distinct Gene Regulatory Networks and Dynamic Developmental Trajectories

Abstract: During early embryogenesis, the transcription factor SOX17 contributes to hepato-pancreato-biliary system formation and vascular-hematopoietic emergence. To better understand Sox17 function in the developing endoderm and endothelium, we developed a dual-color temporal lineage-tracing strategy in mice combined with single-cell RNA sequencing to analyze 6,934 cells from Sox17-expressing lineages at embryonic day 9.0-9.5. Our analyses showed 19 distinct cellular clusters combined from all three germ layers. Diffe… Show more

“…Forward programming has proven to be an effective strategy for the generation of various somatic cell types ( Sugimura et al., 2017 ). SOX17 plays a pivotal role in governing the specification of the endothelial lineage and endothelial regeneration ( Goveia et al., 2014 ; Liu et al., 2019 ; Trinh et al., 2023 ). Furthermore, SOX17 expression has been observed in many other cells, such as definitive endoderm cells ( Jiang et al., 2021 ) and primordial germ cells ( Irie et al., 2015 ).…”
