Siramesine causes preferential apoptosis of mast cells in skin biopsies from psoriatic lesions

Abstract: SummaryBackground Skin mast cells are implicated as detrimental effector cells in various inflammatory skin diseases such as contact eczema, atopic dermatitis and psoriasis. Selective reduction of cutaneous mast cells, e.g. by inducing targeted apoptosis, might prove a rational and efficient therapeutic strategy in dermatoses negatively influenced by mast cells. Objectives The objective of the present study was to evaluate whether a lysosomotropic agent such as siramesine can cause apoptosis of mast cells pres… Show more

“…Anatomically, MCs are most prominently located perivascularly, in close proximity to nerve fibres, and around HFs. MCs may indeed play a more important role in psoriasis than is widely appreciated . Although it has been reported that the number of MCs is elevated in pruritic psoriatic lesions, one study has shown no difference in plasma histamine levels between psoriasis patients with or without pruritus; this might explain why <20% of itchy psoriasis patients respond to oral antihistamines .…”

“…Skin resident mast cells have a detrimental effect on different inflammatory skin diseases including psoriasis [63,64] and hence, selective reduction in the number or activity of mast cells has been proposed to overcome the symptoms of psoriasis. Mast cells release a myriad of primary and secondary inflammatory mediators [65].…”

“…Psoriasis is a complex immune‐mediated disease affecting 2–4% of the population in Western countries. Apart from its negative influence on the skin and joints, the disease is associated with numerous comorbidities and has a detrimental effect on quality of life . Mast cells are known to participate in initiation and maintenance of chronic inflammatory responses such as psoriasis .…”

“…Continuous expression of E‐selectin in the synovium maintains the influx of circulating inflammatory cells into the joints, which may contribute to synovial hyperplasia, leading to an increased number of mast cells. Siramesine, a lysosomotropic drug, was recently studied by Hagforsen et al . on ex vivo psoriatic skin.…”
