SIRT1 negatively regulates amyloid-beta-induced inflammation via the NF-κB pathway

Abstract: Chronic inflammation induced by amyloid-beta (Aβ) plays a key role in the development of age-related macular degeneration (AMD), and matrix metalloproteinase-9 (MMP-9), interleukin (IL)-6, and IL-8 may be associated with chronic inflammation in AMD. Sirtuin 1 (SIRT1) regulates inflammation via inhibition of nuclear factor-kappa B (NF-κB) signaling, and resveratrol has been reported to prevent Aβ-induced retinal degeneration; therefore, we investigated whether this action was mediated via activation of SIRT1 si… Show more

“…SIRT1 is a NAD(+)‐dependent deacetylase and a critical regulator in various metabolic processes. SIRT1 can directly interact with RelA/P65, subunits of NF‐κB, and inhibit NF‐κB by deacetylating RelA/P65 at lysine 310, leading to the IκBa‐dependent nuclear export of NF‐κB . In addition to the SIRT1/ NFκB pathway, Atr also influences microglial activation.…”

“…Chronic inflammation caused by Ab has been widely reported. 52 RelA is a subunit of nuclear factorkappa B (NF-jB), a key regulator of the inflammation response. 53 The IjB kinase inhibitor of jB kinase epsilon (IKBKE) is an IjB kinase family member that can also activate the AKT and NF-jB pathways; IKBKE is also involved in the interleukin-1 (IL-1) signaling pathway.…”

“…They were first discovered in yeast as a transcriptional repressor called silent information regulator 2 protein (Sir2) (Klar et al , 1979; Rine and Herskowitz, 1987). All eukaryotes encode sirtuins in their genomes as sirtuins have important functions in regulating cellular stress responses (Brunet et al , 2004; Dai et al , 2014; Maksin-Matveev et al , 2015), mitochondrial biogenesis (Brenmoehl and Hoeflich, 2013), apoptosis (Alcendor et al , 2004; Motta et al , 2004), metabolism (Purushotham et al , 2009), fatty acid oxidation (Hirschey et al , 2010), and inflammation (Cao et al , 2013; Liu and McCall, 2013). …”
