Smurf2 Regulates the Senescence Response and Suppresses Tumorigenesis in Mice

Abstract: The E3 ubiquitin ligase Smurf2 mediates ubiquitination and degradation of several protein targets involved in tumorigenesis and induces senescence in human cells. However, the functional role of Smurf2 in tumorigenesis has not been fully evaluated. In this study, we generated a mouse model of Smurf2 deficiency to characterize the function of this E3 ligase in tumorigenesis. Smurf2 deficiency attenuated p16 expression and impaired the senescence response of primary mouse embryonic fibroblasts. In support of a f… Show more

“…Nonetheless, our and subsequently other studies provided strong evidence on tumor suppressor functions of Smurf2 [162][163][164]. We demonstrated that Smurf2 −/− mice are prone to development of a wide spectrum of tumors in different organs and tissues, including hepatocellular, lung and mammary carcinomas, as well as lymphomas [162].…”

“…We utilized a novel spontaneously producing DLBCL mouse model known as a Smurf2-deficient mice (Smurf2 T/T , T for the gene-trapping allele) described in detail in previous publications [32, 33]. Age matched C57BL/6 mice were used as wild-type mice and controls.…”

“…Age matched C57BL/6 mice were used as wild-type mice and controls. Spleen tissue and bone were harvested using previously described methodology after mice were sacrificed from five Smurf2 T/T and wild-type two month old mice [32, 33]. 100 to 200 μl of peripheral blood was harvested from the mice using facial vein blood collection.…”

“…Adventitious expression of Smurf2 is sufficient to induce senescence in early passage cells (Zhang & Cohen, 2004; Ramkumar et al ., 2012), while deficiency in Smurf2 expression impairs the senescence response in culture and in vivo (Kong et al ., 2011; Ramkumar et al ., 2012). In this study, we found that Smurf2 deficiency led to increased proliferation and an expanded HSC compartment in bone marrow.…”
