Social stress‐potentiated methamphetamine seeking

Abstract: Human studies of substance use disorder show that psychological stress and drug availability interact following rehabilitation, contributing to the high relapse potential. Social stressors trigger particularly strong motivation for drug, but how this affects neuronal function to increase relapse is unknown. Animal models, which allow for the dissection of neural mechanisms, primarily utilize physical stressors to trigger relapse. To recapitulate psychosocial post-rehabilitation challenges in animals, we develo… Show more

“…Less is known about the stagesetting effects of stress in other drug classes. Blouin et al (2019) have found that social defeat stress potentiates methamphetamineinduced reinstatement and the corresponding Fos response in the basolateral amygdala following self-administration in rats. However, the degree to which stress can potentiate drug seeking with other classes of drugs and the degree to which there is circuit and mechanistic overlap awaits further investigation.…”

“…Indeed, unresolved PTSD is largely considered to be a chronic stress disorder [ 103 ]. These studies are paralleled by pre-clinical studies in rodents reporting that stress triggers relapse to Meth and increases drug-seeking [ 8 , 104 ].…”

“…Stress is a well-documented risk factor for the development of addiction and the vulnerability to relapse. The effects of stress also influence patterns of alcohol and Meth abuse [ 8 ] and can further contribute to a triad of Meth, alcohol abuse, and stress.…”

“…Using the SA paradigm, it has been demonstrated that exposure to social stress increases the reinforcing effects of drugs of abuse[ 136 - 139 ]. Recently, resilience to these effects has also been studied using different types of social stress and drugs of abuse, such as cocaine[ 140 - 143 ], methamphetamine[ 144 ] and alcohol[ 145 - 147 ].…”

“…Interestingly, the defeat latency during the episode of SD correlated with reinstatement values and c-Fos-immunoreactivity in the basolateral amygdala; priming-induced reinstatement and c-Fos were both potentiated in rapidly defeated rats. Conversely, these effects were not observed in rats that were undefeated during the social encounter, but inactivation of the basolateral amygdala induced potentiation of reinstatement, suggesting that this structure mediates resilience against SD stress[ 144 ]. The positive correlation between reinstatement and passive coping (high values of reinstatement in animals with lower latency defeat) was reported in another study, although the authors questioned its real meaning, and proposed that active coping behaviors during SD episodes were associated with the magnitude of reinstatement[ 257 ].…”
