Soft Polydimethylsiloxane-Supported Lipid Bilayers for Studying T Cell Interactions

Abstract: Much of what we know about the early stages of T cell activation has been obtained from studies of T cells interacting with glass-supported lipid bilayers that favor imaging but are orders of magnitude stiffer than typical cells. We developed a method for attaching lipid bilayers to polydimethylsiloxane polymer supports, producing ''soft bilayers'' with physiological levels of mechanical resistance (Young's modulus of 4 kPa). Comparisons of T cell behavior on soft and glass-supported bilayers revealed that whe… Show more

“…In this hypothetical setup, ICAM-1 is immobilized on a hexagonal gold nanoparticle array with defined interparticle spacing and pMHC is embedded onto a fluid bilayer ( 55 ). Further enhancing physiological relevance, the SLB could be deposited onto soft polydimethylsiloxane substrates mimicking the physiological relevant stiffnesses of APCs ( 56 ). These proposed experimental systems will enable a systemic investigation on how the immobilized pool of ICAM-1 alters the ability of naïve cells to discriminate antigens, T cell adhesion, TCR signaling, and actin dynamics.…”

“…Although glass is the most commonly used substrate, poly(dimethylsiloxane) (PDMS) is also widely used owing to its unique features. PDMS can be easily customized via soft lithography, and thus it is a versatile platform for forming lipid bilayers with complex geometries. Moreover, the controllable mechanical properties of PDMS can be tailored for studying various membrane features with PDMS-supported lipid bilayers. In addition, PDMS-supported lipid bilayers are stable up to 132 h after preparation and hence are appropriate support for the above membrane applications.…”

“…Thus, mechanical stress from stiffness-based membrane deformation acts as a rheostat, leading to differential expression of genes involved in TCR/CD3 induced T cell functions 12 , 199 201 . However, pMHC and ICAM-1 on SLBs over soft (4 kPa) polydimethylsiloxane supports triggered early TCR signaling similarly to SLBs on stiff (GPa) glass supports 202 .…”
