Somatostatin receptor 5 and cannabinoid receptor 1 activation inhibit secretion of glucose-dependent insulinotropic polypeptide from intestinal K cells in rodents

Abstract: Aims/hypothesisGlucose-dependent insulinotropic polypeptide (GIP) is an enteroendocrine hormone that promotes storage of glucose and fat. Its secretion from intestinal K cells is triggered by nutrient ingestion and is modulated by intracellular cAMP. In view of the proadipogenic actions of GIP, this study aimed to identify pathways in K cells that lower cAMP levels and GIP secretion.MethodsMurine K cells purified by flow cytometry were analysed for expression of Gαi-coupled receptors by transcriptomic microarr… Show more

“…Five somatostatin receptors (SSTRs) have been identified, all thought to be G i -coupled, leading to inhibition of adenylyl cyclase and a reduction in intracellular cAMP levels. In GLUTag and primary L cells, somatostatin is believed to inhibit GLP1 and PYY release via SSTR5 (94). Ghrelin secretion is also inhibited by somatostatin, potentially involving SSTR1, -2, and -3 (48).…”

“…Octreotide suppressed not only plasma GLP-1 but also GIP concentration within 15 minutes during 75 g-OGTT. Somatostatin receptors exist as five isoforms (SSTR1-SSTR5), of which sst5 is expressed by the rat L-cell and the rodent K-cell [18,19]. Somatostatin inhibits GIP and GLP-1 secretion from small intestinal cultures through SSTR5 [17].…”

“…Previous studies employing several experimental models including single K-and L-cells have shown that the hormones are secreted in response, for example, to glucose, which after entry via the sodium-glucose transporter-1 results in membrane depolarization and exocytosis secondary to calcium influx in both cell types (Reimann et al 2008, Parker et al 2009. Other studies have shown that stimuli activating the cAMP system may also result in both GLP1 and GIP secretion (Simpson et al 2007, Moss et al 2012, Hauge et al 2014, Svendsen et al 2015.…”
