Sorafenib-loaded hydroxyethyl starch-TG100-115 micelles for the treatment of liver cancer based on synergistic treatment

Abstract: Tumor microenvironment is closely related to the occurrence and development of liver cancer. Tumorassociated macrophages (TAMs) are an important part of tumor microenvironment promoting tumor deterioration and metastasis by inhibiting immune cells. Previous studies showed that PI3Kc inhibitor could reverse the phenotype of TAMs, relieve immunosuppression and sensitize chemotherapy drugs, suggesting that the combination of PI3Kc inhibitor and chemotherapeutics is likely to bring new breakthroughs in the treatme… Show more

“…The synthesis of the TC‒mPEG‒PLGA copolymer was accomplished by reacting the carboxyl moiety of mPEG in DDC with the amino group of TC. The TC‒mPEG‒PLGA copolymer was an amphiphilic compound, wherein mPEG acted as hydrophilic groups, while PLGA served as hydrophobic groups ( Li and Zhao, 2019 ). The TC‒mPEG‒PLGA self-assembled into micelles in water on account of the hydrophobic segments segregating from water to form an inner core and the hydrophilic segments orientating outward to contact with water.…”

“…Phosphoinositide 3-kinase gamma (PI3Kγ) regulates the immunosuppressive properties of TAMs. PI3Kγ inhibitors, such as TG100-115, can induce the expression of MHC-II and proinflammatory cytokines, skewing macrophages to the M1 phenotype while reducing immunosuppressive molecules and inhibiting tumor progression [170].…”

“…After 95 h, cumulative Cur release has higher at pH = 5.5 (23%) than at pH = 7.4 (8%). In a similar study, PEG 5000 and TG100-115, which is an exclusive PI3Kc inhibitor, were conjugated on hydroxyethyl starch in order to obtain sorafenid-loaded micelles by a dialysis method [ 97 ]. The drug release was investigated as a function of pH (5.5, 6.8, and 7.4), and it appeared that the higher release rate was observed at low pH value.…”
