Spirobisnaphthalenes effectively inhibit carbonic anhydrase

Abstract: This study explores the correlation between human carbonic anhydrase (CA, EC 4.2.1.1) isoforms I and II (hCA I, II) and the inhibitory features of some spirobisnaphthalene derivatives. A group of spirobisnaphthalenes was synthesized and their hCA I and II inhibitory effects was investigated. The K i values were similar for both CA isoenzymes, the compounds showing good inhibitory activity. K i values ranged between 1.60 and 460.42 mM for hCA I and between 0.39 and 419.42 mM for hCA II, respectively. The spirob… Show more

“…In mammals, CA II, which generally exists in red blood cells in lower concentrations, has approximately 10 times higher activity compare with CA I [99][100][101][102] . Cytosolic hCA I isoenzyme is ubiquitously expressed in body, and available in high concentrations in blood and gastrointestinal tract.…”

“…[1][2][3] Carbonic anhydrase (CA) is ubiquitous metalloenzyme that catalyze the two physiologically important processes: the first, the hydration of carbon dioxide to bicarbonate ion and a proton, and the second, the dehydration of bicarbonate ion. [4][5][6] These simple reactions provide acid-base equilibrium and ion equilibrium, so carbonic anhydrases are involved in many physiological processes such as pH regulation, ion transport, electrolyte secretion, calcification, and some biosynthetic reactions. 4,7 Among the mammalian carbonic anhydrases, α-carbonic anhydrases, sixteen different CA isoforms are expressed in humans.…”
