Sporogony and sporozoite rates of avian malaria parasites in wild Culex pipiens pallens and C. inatomii in Japan

Abstract: BackgroundMalaria infection in mosquitoes is traditionally detected by microscopic examination for Plasmodium oocysts and sporozoites. Although PCR is now widely used, the presence of parasite DNA in a mosquito does not prove that sporogony is achieved. Thus, detection of sporozoites by microscopy is still required to definitively identify vector mosquitoes. The aim of this study was to confirm sporogony of avian Plasmodium spp. in Culex pipiens pallens and C. inatomii caught from the wild.FindingsMosquitoes c… Show more

“…Both sporozoites and oocysts of CXPIP09 have been confirmed in Cx. pipiens group, verifying that this mosquito species is a competent vector [ 5 ]. This lineage was detected from mosquitoes in all sampling years, strongly suggesting that this lineage is maintained at the study site.…”

“…As part of conservation efforts, ex-situ populations have been established in zoos, aquariums and other breeding facilities throughout the world. Avian malaria, caused by Plasmodium parasites, is mainly transmitted by Culex mosquitoes [ 2 – 5 ]. Due to their evolutionary background in environments of little contact with vector species, penguins are mostly naïve to avian malaria and many lethal cases of avian malarial infections have been reported in captive penguins [ 6 , 7 ].…”

“…pseudotitillans , suggesting that these species are potential vectors of avian malaria parasites in the SDTF of Brazil. However, detection of Plasmodium DNA using molecular methods alone does not confirm transmission; one must also visualize sporozoites in the salivary glands of mosquitoes using microscopy or should conduct experimental infections to confirm true vector identity [38, 39]. Nevertheless, our study represents an important step in identifying vectors of avian malaria parasites in the Neotropics.…”

“…Avian malaria vectors can have negative thoraces when corresponding abdomens are positive in experimental and natural conditions [39, 45, 49]. For example, detection of positive thoraces by PCR starts at nine days after experimental infections of competent vectors, with positivity rates varying from 22.2 to 60% afterwards [45].…”

“…Species in the Culex pipiens complex are world widely distributed [4,30,31], meanwhile important vectors to plenty of virus and parasites, including West Nile virus, Wuchereria bancrofti and Diro laria immitis [29,32,33]. Culex pipiens pallens, commonly found north of 37°N in China, Japan and Korea, is one of the major mosquito species natively responsible for biting human and disease transmission [34].…”
