Spread of extended-spectrum beta-lactamase-producing Escherichia coli from a swine farm to the receiving river

Abstract: The dissemination of drug-resistant bacteria into different environments has posed a grave threat to public health, but data on the spread of extended-spectrum beta-lactamase (ESBL)-producing Escherichia coli (E. coli) from animal farms to the receiving river are limited. Here, 57 ESBL-producing E. coli isolated from a pig farm and the receiving river were analyzed in terms of drug resistance, ESBL genes, and enterobacterial repetitive intergenic consensus (ERIC). The results showed that ESBL-producing E. coli… Show more

“…The total volume of PCR reaction was 25 μ L, including 10x buffer 2.5 μ L, 2.5 mmol/L dNTP 2.0 μ L, forward and reverse primers (20 pmol/L) 1 μ L for each, 25 mmol/L MgCl 2 1.5 μ L, Taq polymerase 5 U, and genome DNA 1.5 μ L. The ERIC-PCR was completed by an initial heat activation of 10 min at 94°C, then 31 cycles of 30 s at 92°C and 60 s at 40°C, 8 min at 65°C, and an extension of 8 min at 65°C. PCR products were run on an agarose gel electrophoresis (2%) at 100 V for 1 h [ 15 ].…”

“…The unweighted pair group method using averages algorithm (UPGMA) was used to obtain the clustering dendrograms (NTSYS-pc 2.10 software). The individual isolates were regarded as an independent operational taxonomic unit (OUT), and isolates with a similarity over 90% were assumed to be of the same origin [ 15 ].…”

“…Date of publication ranged from 1995 to 2016, with the highest number of publications in 2015 ( n = 11). The spatial scale for the sampling frame ranged from 40 m (Li, Song, et al, ) to 450 km (Sulzner et al., ); six studies did not report a specific distance.…”

“…Furthermore, the ESBL/AmpC-producing E. coli in animal farms could influence public health through environmental pollution (Li et al, 2015). Fecal emission and the airborne route might be possible ways of ESBL/AmpC emission into the environment (Yuan et al, 2010; Friese et al, 2013; Laube et al, 2014).…”
