Molecular oxygen, a basic component of aerobic metabolism, is an inevitable component of all living organisms. However, a portion of the oxygen used by the cells is converted to several harmful reactive oxygen species (ROS) and free radicals. In low/moderate concentrations, free radicals serve as beneficial compounds and are involved in normal physiological functions. Excessive free radical generation results in oxidative stress, with the prime targets being the fundamental building blocks of cell, such as lipids, protein, and DNA. Oxidative damage to these molecules leads to growth arrest, senescence, or apoptosis which play crucial role in the development of several pathological conditions of humans. A growing body of recent literature suggests that scavenging these ROS alleviates the diseases completely. The documented literature suggests that many herbal extracts possess antioxidant effects. Among them, brahmi (Bacopa monneiri) has received increasing attention as a traditional medicine for treatment of several disorders. Previous research has shown that Bacopa monneiri is used to cure various ailments such as cardiovascular diseases, cancer, neurodegenerative diseases, and diabetes, among others. The present review focuses on the therapeutic potential of Bacopa monneiri in the management of diseases that are caused by oxidative stress and ROS by acting as a free radical scavenger.

The convolution associated with memory is being resolved with advancement in neuroscience. According to the concurrent assumptions, synaptic plasticity forms one of the basis of memory formation, stabilization and strengthening. In Alzheimer's disease (AD), which is generally characterized by memory dysfunction, connections amongst the cells in the brain are attenuated or lost leading to degeneration of neural networks. Numerous attempts have been made to find new therapies for memory dysfunction with increasing attention and investments being laid on herbal drugs. Many herbal plants and extracts have already documented beneficial results when tested for antiamnesic effects. Brahmi (Bacopa monniera) is one such common herbal drug, which is employed for a long time in the Indian and Chinese medical system in order to treat several disorders. Previous research has shown that Brahmi exerts many pharmacological effects including memory boosting capacity in the treatment of Alzheimer's disease and Schizophrenia, exhibiting antiparkinsonian, antistroke, and anticonvulsant potentials. The present review discusses the chemical constituents of Brahmi along with in vitro and in vivo studies based on the pharmacological effects exerted by it. The efficacy of Brahmi in treating various disorders has evoked sufficient research in recent years and now it is a time to launch multiple clinical trials.

Stability Testing of Herbal Drugs: Challenges, Regulatory Compliance and Perspectives

Stability testing is an important component of herbal drugs and products (HDPs) development process. Drugs regulatory agencies across the globe have recommended guidelines for the conduct of stability studies on HDPs, which require that stability data should be included in the product registration dossier. From the scientific viewpoint, numerous chemical constituents in an herbal drug are liable to varied chemical reactions under the influence of different conditions during its shelf life. These reactions can lead to altered chemical composition of HDP and consequently altered therapeutic profile. Many reports on stability testing of HDPs have appeared in literature since the last 10 years. A review of these reports reveals that there is wide variability in temperature (-80 to 100 °C), humidity (0-100%) and duration (a few hours-36 months) for stability assessment of HDPs. Of these, only 1% studies are conducted in compliance with the regulatory guidelines for stability testing. The present review is aimed at compiling all stability testing reports, understanding key challenges in stability testing of HDPs and suggesting possible solutions for these. The key challenges are classified as chemical complexity and biochemical composition variability in raw material, selection of marker(s) and influences of enzymes.
