Star-Shaped Polylactide Dipyridamole Conjugated to 5-Fluorouracil and 4-Piperidinopiperidine Nanocarriers for Bioimaging and Dual Drug Delivery in Cancer Cells

Abstract: Star-shaped polylactide (SSPLA) nanoparticles (NPs) with dipyridamole (DIP) core conjugated with 5-fluorouracil (5FU) and 4-piperidinopiperidine (4PIP) were designed and synthesized to achieve apoptosis by synergistic dose-dependent delivery than using free drugs. In our current investigation, DIP was employed as the initiator in ring-opening polymerization reaction to make SSPLADIP. We then described the synthesis of tailor-made, self-assembled, carboxyl group-substituted fluorescent SSPLADIP conjugated with … Show more

“…For example, Abulateefeh and coworkers [ 58 ] reported a good fit to zero-order release kinetics for PLA microcapsules loaded with fluorescein sodium. Similarly, Moorkoth and coworkers [ 59 ] applied this equation to the administration of different anticancer drugs through PLLA nanoparticles.…”

“…These bioimaging applications can be combined with therapeutic applications, such as drug delivery and PTT, to simultaneously perform treatment and diagnosis to enable precision medicine that can be tailored to each patient ( Moorkoth et al, 2021 ; Yang et al, 2021 ). Nanoscale particles can be targeted to desired positions through surface functionality ( Wijetunga et al, 2020 ).…”

“…For the past 70 decades, fluoropyrimidine 5-fluorouracil (5FU) has been widely utilized as a frontline medication for the treatment of a wide variety of tumors, including colon, cervical, lung, and breast cancers. When given intravenously, it is aqueous soluble and stable (De Luca et al, 2016 ; Ai et al, 2019 ; Dai et al, 2020 ; Moorkoth et al, 2021 ). However, for early colorectal cancer, 5FU has a response rate ∼15%, and its biocompatibility decreases life times.…”
