Starch Digestion–Related Amylase Genetic Variant Affects 2-Year Changes in Adiposity in Response to Weight-Loss Diets: The POUNDS Lost Trial

Abstract: Salivary and pancreatic amylases (encoded by AMY1 and AMY2 genes, respectively) are responsible for digesting starchy foods. AMY1 and AMY2 show copy number variations that affect differences in amylase amount and activity, and AMY1 copies have been associated with adiposity. We investigated whether genetic variants determining amylase gene copies are associated with 2-year changes in adiposity among 692 overweight and obese individuals who were randomly assigned to diets varying in macronutrient content. We fo… Show more

“…Individuals with lower AMY1 CNV and therefore low levels of salivary amylase, may be less well adapted to the digestive demands of a modern high-starch diets, with more pronounced postprandial glycemic excursions, impaired glucose tolerance and subsequent development of insulin resistance [32]. Consistent with this hypothesis, in one study on overweight and obese individuals, a high AMY1 CNV was associated with lower fasting blood glucose levels [77].…”

“…In total, seven publications assessed the interaction of genotype and protein intake on weight loss [39,40,42,43,46,49,51] (Table 4). All of them analyzed data from the POUNDS Lost trial and used an additive model for genetic analysis.…”

“…Only three individuals carried the rs12913832-G allele associated with blue eye colour, seven had the A allele associated with brown eye colour, and eight had both alleles. The minor A allele of rs11185098 is positively associated with AMY1 (amylase 1) gene copies and high amylase activity, responsible for starch digestion 27 . Only one individual was found to be homozygous for the G allele and six had both alleles, while no homozygous carrier for the A allele was found.…”
