Stem Cell Factor Combined with Matrix Proteins Regulates the Attachment and Migration of Melanocyte Precursors of Human Hair Follicles in Vitro

Abstract: In conjunction with matrix proteins, stem cell factor (SCF) plays an important role in the migration of melanocyte precursors (MPs) derived from the mouse embryo. However, no studies have demonstrated an effect of SCF on human follicular MPs migration in vitro. In this report, first we demonstrate the immature state of the follicular MPs. Then cell attachment rate was measured by 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide method. Standard 48-well chemotaxis chambers were used for a transfilte… Show more

“…To the best of our knowledge, our work is the first to use the skin samples combined with SHF-DPCs samples to explore novel hub genes associated with the growth and development of SHF. Consistent with published data, the enrichment of these DEGs in several GO terms, such as extracellular structure organization, cell chemotaxis, cellular component, collagen-containing extracellular matrix, and collagen binding, confirms their involvement in the growth and development of HF [39][40][41][42]. Enrichment of the identified DEGs in some KEGG pathways, such as Staphylococcus aureus infection, phenylalanine metabolism, and adipocytokine-signaling pathway, also suggests their relevance in the growth and development of HF.…”

“…The remodeling of the actin cytoskeleton has been considered as a necessary process for the migration of different cell types, including melanocytes ( 19 , 20 ). Therefore, the effects of ZMIZ1 on actin remodeling in the cytoskeleton were investigated using immunocytochemistry.…”

“…Endothelin-1 promotes melanocyte dendricity [16] and regulates the differentiation, growth, and survival of melanocytes via the microphthalmia-associated transcription factor-regulated GPNMB pathway [17]. The stem cell factor (SCF)/c-kit pathway mediates epidermal cell adhesion, migration, and proliferation of melanocyte stem cells [18]. Basic fibroblast growth factor induces melanocyte migration through increased expression of the focal adhesion kinase p125 FAK [19].…”
