Stoichiometric limitations in assembly of active recombinant retrovirus

Abstract: Although recombinant retroviruses are widely used in gene therapy and as gene transfer vehicles for basic biological studies, their titers are very low as compared to other recombinant viral systems, e.g., adenovirus. We investigated the rate-limiting steps in production of LacZ-encoding ecotropic (CRE BAG 2) and amphotropic (Psi-CRIP) retrovirus. We found that ecotropic retrovirus producer cells produced a large number of inactive viral particles because they were severely limited by the amount of mRNA that w… Show more

“…The next day, the medium was replaced with 30mL of DMEM+10%FBS containing sodium butyrate (10mM) that was previously shown to improve virus production [43]. Two days later, the supernatant was harvested, filtered, ultra-centrifuged at 50,000g for 2 hr and the lentiviral pellet was dissolved by 300μL fresh medium (DMEM+10%FBS) to yield 100x concentrated lentivirus stock.…”

“…The absence of one of these properties results in the production of deficient, noninfectious vectors. The packaging cell lines currently used to produce RVs are known to yield low infectious titers (10 6 -10 7 infectious particle (IP)/mL) and heterogeneous RVs populations lacking one or more of the elements mentioned above [5]. In addition, RVs biological activity is intrinsically very susceptible to environmental conditions, mainly temperature [6,7].…”
