Streptomyces Levis ABRIINW111 Inhibits SW480 Cells Growth by Apoptosis Induction

Abstract: Purpose: Streptomyces sp., a dominant genus in Actinomycetes, is the source of a wide variety of secondary metabolites. Microbial metabolites can be utilized as novel anticancer agents; with fewer side effects. The present article illustrated the anti-carcinogenic effect of the ether extracted organic metabolites derived from Streptomyces bacteria on SW480 colon cancer cell line. Methods: MTT assay was performed in order to investigate the cytotoxicity effect of metabolites on SW480 cells. Apoptosis and cell cy… Show more

“…Their presence is very important in the prevention of colonisation of more virulent micro-organisms [155] Streptococcus gordonii May have protective role, such that epithelial cells do not respond to Porphyromonas gingivalis-induced epithelial cell proliferation stimulation [156] Streptomyces Some species have been shown to inhibit cell growth [157], including apoptosis of cancer cell lines [158] Neisseria Can break down toxins of tobacco…”

“…Cells were washed and stained with PI (5 μL/1 × 10 6 ) and immediately analyzed by flow cytometry. 20 In more detail, intracellular staining was performed for caspase-3. Cells were washed first with PBS, fixed with 4% formaldehyde for 30 min, resuspended in 0.5 mL of the Permeabilization solution, incubated for 20 min, washed with PBS, and incubated with anti-caspase-3 antibodies for 20 min.…”

“…Ether extract of Streptomyces levis ABRIINW111 showed an increase in p53 expression, cell cycle arrest in G1 and G2/M phases and apoptosis in SW480 colon cancer cell line (Faramarzian et al 2018 ). Metabolites from this extract decreased anti-apoptotic K-RAS and Bcl-2 proteins expression and increased pro-apoptotic p53 protein expression (Fatourachi et al 2018 ).…”

“…Metabolites derived from Streptomyces sp. SY-103 have been shown to induce apoptosis in human leukemic cells upon treatment; through activating caspase-3 and inactivating AKT (Faramarzian et al 2018 ). Some of these metabolites are pure cytotoxic compound (PCC), spicamycin and its derivative KRN5500, which were found to induce apoptosis by decreasing expression of Bcl-2 and modulating the normal localization pattern of promyelocytic leukemia protein (PML) from speckled pattern to microspeckled pattern in myeloid and lymphoid leukemic cells (Jeong et al 2010 ; Zhang et al 2000 ).…”
