Structural Characteristics of Rehmannia glutinosa Polysaccharides Treated Using Different Decolorization Processes and Their Antioxidant Effects in Intestinal Epithelial Cells

Abstract: Polysaccharide decolorization is a key determinant of polysaccharide structure. In this study, two purified Rehmannia glutinosa polysaccharides, RGP−1−A and RGP−2−A, were obtained after decolorization using the AB-8 macroporous resin and H2O2, respectively. RGP−1−A (molecular weight (Mw) = 18,964 Da) and RGP−2−A (Mw = 3305 Da) were acidic and neutral heteropolysaccharides, respectively, and were both polycrystalline in structure. FTIR analysis revealed that RGP−1−A was a sulfate polysaccharide, while RGP−2−A h… Show more

“…Here, we reviewed some human monoculture models used to study the consequences of nutrients, toxicants or drugs in terms of their absorption or interaction with the intestinal epithelial barrier. Although we restricted this review of the literature to human cell lines, there also exist murine and porcine cell lines [ 81 , 82 , 83 , 84 ]. The principal disadvantage of monoculture models is that they do not reflect the diversity of the mucosa with its several cell types and multiple functions.…”

“…Two purified polysaccharides (RGP-1-A and RGP-2-A) were obtained from Rehmannia glutinosa after decolorization using AB-8 macroporous resin and H 2 O 2 respectively, and their M w values were 18.964 and 3.305 kDa. RGP-1-A showed significantly higher antioxidant capacity on H 2 O 2 -induced IPEC-1 cells and caused more up-regulation on mRNA expressions of Nrf2, HO-1 and NQO1 and less Keap1 mRNA expression ( 208 ).…”

“…Two purified polysaccharides (RGP-1-A and RGP-2-A) obtained from Rehmannia glutinosa were determined to have GalA contents of 19.02 and 1.1%. RGP-1-A showed significantly better cytoprotection on H 2 O 2 -induced IPEC-1 cells and caused observably more increments in mRNA expressions of Nrf2, HO-1 and NQO1 and reduction in Keap1 mRNA expression ( 208 ).…”

“…Polysaccharides—biopolymers composed of several monosaccharides connected by glycosidic bonds—are the main active functional components of RG. Polysaccharides are typically extracted and purified from the roots and stems of RG and have a relative molecular weight of 4 × 10 3 to 5 × 10 4 Da [ 19 , 20 ]. Owing to their excellent antioxidant [ 20 ], anti-inflammatory [ 15 ], and immune-enhancing [ 21 ] properties, polysaccharides from RG (RGP) have attracted immense attention.…”
