Acacia tortuosa produces a clear gum, very soluble in water. Previous reports showed that it was constituted by four fractions, one of them an arabinogalactan-protein complex. The elucidation of the A. tortuosa gum structure by the combination of classical chemical methods, size exclusion chromatography and NMR spectroscopy, was the objective of this investigation. The data obtained show that the heteropolysaccharide is an arabinogalactan type II, highly ramified, with lateral chains at C-2 as well as at C-6 of the galactose 3-O residues; mono-O-substituted galactoses were not detected. There are residues of mannose, the arabinose, pyranose predominantly, is terminal and 2-O-linked. The abundance of the 4-O-methyl-α-d-glucuronic acid was not previously reported. The proteic fraction is probably represented by an arabinogalactan-protein complex that binds poorly with β-glucosyl Yariv reagent, and two glycoproteins. The NMR spectra suggest that the carbohydrate links to hydroxyproline through the galactose (galactosylation).

RESUMO -Spondias dulcis Forst. F. vem recebendo atenção especial por secretar uma goma que possui ação medicinal. Em outras espécies de Anacardiaceae, a maioria dos estudos sobre as estruturas secretoras referese à ultra-estrutura e desenvolvimento dos ductos, existindo poucas informações quanto à natureza química do secretado. O objetivo do presente trabalho foi caracterizar estrutural e histoquimicamente o sistema secretor do caule de S. dulcis. Amostras do caule jovem de plantas com 45 dias de idade foram submetidas às técnicas usuais em anatomia vegetal e a testes histoquímicos. O sistema secretor de S. dulcis apresenta ductos e idioblastos. Os ductos, muito longos, encontram-se associados ao floema e na medula, enquanto os idioblastos ocorrem no córtex e, também, associados ao floema. Os ductos apresentam um epitélio secretor unisseriado, formado por células cuneiformes a tabulares, de paredes delgadas, citoplasma denso e núcleo volumoso com nucléolo proeminente. Nos ductos associados ao floema, observam-se, externamente ao epitélio, duas a três camadas de células parenquimáticas, achatadas e dispostas radialmente, formando uma bainha. Os testes histoquímicos evidenciaram a presença de taninos nos idioblastos corticais. A secreção dos ductos é constituída, em sua maioria, por óleos essenciais, polissacarídeos e compostos fenólicos (provavelmente agliconas flavonóicas), o que permite caracterizá-la como uma gomorresina.Palavras-chave: Anacardiaceae, Spondias dulcis, anatomia caulinar, ductos secretores, idioblastos secretores e histoquímica. Degradation by-products of ancient paper leaves from wash waters

International audienceWater cleaning of paper leaves is a preliminary step in many customary deacidification treatments of ancient books in conservation workshops. Usually, the washing solutions are considered “waste waters” and they are thrown away. In this work, an analytical protocol for the analysis of wash waters for the assessment of the conservation state of ancient books is proposed. Wash waters of leaves under different conservation conditions of a 16th-century- rinted book were investigated using UV-Vis, IR, Raman, NMR, EPR, XRF, ESI-MS, ICP-MS, CE-PDA and elemental microanalysis. Analysis of the wash water extracts allowed one to identify a large number of degradation by-products, ranging from low-molecular mass organic acids to cellulose oligomers. Reasons for the very different conservation conditions of the leaves emerged, indicating that wash water analysis provides a deep insight into the conservation state and composition of leaves as well as degradation reactions occurred on the
