Structural Investigation of Cell Wall Xylan Polysaccharides from the Leaves of Algerian Argania spinosa

Abstract: Xylan-type polysaccharides were isolated from the leaves of Argania spinosa (L.) Skeels collected in the Tindouf area (southwestern Algeria). Xylan fractions were obtained by sequential alkaline extractions and purified on Sepharose CL-4B. The xylan structure was investigated by enzymatic hydrolysis with an endo-β(1→4)-xylanase followed by chromatography of the resulting fragments on Biogel P2, characterization by sugar analysis and matrix-assisted laser desorption ionization-time of flight mass spectrometry (… Show more

“…The peak intensity ratio between the band at 1402 cm -1 (υ (C-O) ) and those at 1554 cm -1 (υ (C=O) ) is larger in the spectrum of the H 2 -KOH4M fraction than the corresponding ratio in the H 1 -KOH1M fraction spectrum. These results confirm the acidic character of all the extracted hemicelluloses 6,27 .…”

“…Furthermore, CO2, the second major component can be sequestered and used to produce chemicals [53]. Hemicellulose can also be found in the primary cell wall in contrast to the cellulose which is present mainly within the secondary wall [44]. It is in turn the second most abundant polysaccharide compound in nature.…”

“…Polysaccharides extracted from A. spinosa L. skeels leaves from Morocco were reported as xylan, made up of a β-(1,4)-D-Xylp main chain, substituted with 4-O-Me-D-GlcpA [138]. Hachem et al described the same xylan structure in polysaccharides extracted from Algerian A. spinosa L. Skeels leaves, but the main chain was also substituted with L-Araf residues, in addition to 4-O-Me-D-GlcpA [139]. Other specific patterns and distributions all along the main backbone were described in the literature, with specific xyloglucan oligosaccharides such as XXXG, XXFG, XLXG/XXLG, XLFG fragments.…”
