Structure and membrane remodeling activity of ESCRT-III helical polymers

Abstract: The Endosomal Sorting Complexes Required for Transport (ESCRT) proteins mediate fundamental membrane remodeling events that require stabilizing negative membrane curvature. These include endosomal intralumenal vesicle formation, HIV budding, nuclear envelope closure and cytokinetic abscission. ESCRT-III subunits perform key roles in these processes by changing conformation and polymerizing into membrane-remodeling filaments. Here, we report the 4 Å resolution cryo-EM reconstruction of a one-start, double-stran… Show more

“…In the autoinhibited dynamin tetramer, an intramolecular contact of the pleckstrin homology (PH) domain with the oligomerization surface of the stalk prevents the assembly in the cytosol (25), whereas an autoinhibitory contact of the C-terminal EH domain with the GTPase domains takes over this function in EHDs. Such intramolecular inhibitory contacts are also observed in other peripheral or integral membrane proteins, such as BAR-domain containing proteins (26,27), SNAREs (28), ESCRT-III (29), or the WASP protein (30) (Fig. S8 B-E).…”

“…5M to P). These partially enveloped structures are exactly those expected to accumulate when neck closure and scission are inhibited by dominant negative Vps4-EQ-GFP (15,59,72,73,76,93). Interestingly, the rim or neck of the unsealed envelope is decorated by a ring of structures resembling beads on a string (Fig.…”

“…It may be that UL36p-null capsids are structurally deficient in ESCRT III/Vps4 recruitment at a level of resolution beyond that used in our studies. Alternatively, UL36p or a UL36p-dependent factor(s) might act very late in envelopment, after Vps4 has been recruited to ESCRT III, to detach it from its membrane anchors (72,93).…”

“…The mechanisms of MV biogenesis still require further study. One accepted mechanism mediated by ESCRT machinery (Figure 4), which is similar to the classic mechanism of Exos biogenesis, showed that MVs could be formed via the recruitment of ESCRT‐III proteins 74, 75, 76. In addition to the ESCRT‐dependent mechanism, it has been hypothesized that there is also an ESCRT‐independent mechanism of MV biogenesis.…”
