Subcellular Localization and Properties of a Cholinergic Receptor Isolated From Housefly Heads

Abstract: —Protein(s) possessing the ligand‐binding properties expected of the insect cholinergic receptor were demonstrated in both aqueous and chloroform‐methanol extracts of an 80,000 g supernatant obtained from frozen housefly heads. There is reasonable correlation in both the properties and concentration of the housefly head cholinergic receptor in these radically different extracts. Binding constants for decamethonium, acetylcholine, nicotine and atropine have been determined with aqueous housefly head extracts us… Show more

“…The 2000 y centrifugation removes eye pigment granules and other debris. As can be seen in Table 2 $ These data are taken from the results of DONNELLAN et al (1975). In their experiments, they prepared the decamethonium binding component by differential centrifugation and measured binding by ultrafiltration.…”

“…The capacity of the four fractions to bind ~-['~C]glutamate in the presence and absence of 1 m-D-glutamate was measured by the ultrafiltration technique described by Donnellan et al (1975). This allowed the amount of L-glutamate bound in the absence of D-glutamate less that bound in the presence of D-glutamate to be designated as specific 566th MEETING, CAMBRIDGE Table 1…”

“…In an attempt to study ACh interactions with the PGL, saturation experiments were carried out, utilizing twelve concentrations of ['*C]ACh at each pressure level. It has been shown that the nicotinic cholinergic receptor, whether extracted and purified as either a proteolipid (Donnellan et al, 1975;Taylor, 197%~) or a protein (Donnellan et al, 1975;Almon and Appel, 1976), has two specific binding components, with K D values of approximately 0.1 and 10 p~. When the data from the saturation experiments with the PGL at each pressure Table 3.…”
