Subgenomic viral DNA species synthesized in simian cells by human and simian adenoviruses

Abstract: DNA synthesized after infection of simian tissue culture cells (BSC-1 or CV-1) with human adenovirus type 2 or 5 or with simian adenovirus 7 was characterized. It was demonstrated that as much as 40% of the virus-specific DNA in nuclei of infected monkey cells consists of subgenomic pieces. No subgenomic viral DNA species were detected in the nuclei of human (HeLa) cells infected with these adenovirus types. Restriction analysis showed that these short viral DNA molecules contain normal amounts of the sequence… Show more

“…Differences in this region of the genome may relate to the early replicative block of EAds in conventional cell lines and the possible complementation of defective early EAd genes by the AdS genes expressed in 293 cells (23). Incomplete adenovirus particles have been noted for several years, and their nature, origin, and significance have been discussed in relation to theories of Ad DNA replication (4,7,12,16,17,20,25). Our observations are similar to those reported in studies of incomplete particles of other Ad types.…”

“…The ∆E3 virus, Ad5rhFLSC, has been described elsewhere [20]. In brief, 2800 base pairs in the E3 region of Adhr404 [21] were replaced with rhFLSC. The mutant virus dl1520 contains an 827-bp deletion in the region encoding the 55kDa protein [22].…”
