Summary of results of serological tests and diagnoses for 4774 cases of various autoimmune bullous diseases consulted to Kurume University

Abstract: The results in this study of 4774 patients with various AIBDs indicate that our immunological diagnostic method is useful for making diagnoses for most patients with AIBD. However, we need further improvements including new immunological techniques to establish more satisfactory methods.

“…Patients with BP show IgG antiepidermal basement membrane zone autoantibodies that react primarily with BP230 and BP180 . While major epitopes for IgG anti‐BP180 autoantibodies reside within the extracellular BP180 NC16a domain, IgG antibodies in BP sera react with various domains on the entire BP230 molecule .…”

“…Indirect immunofluorescence detected IgG anti‐BMZ autoantibodies at a titre of 1:10, located in both the epidermal and dermal sides of the salt split skin. An immunoblot (IB) study was performed according to the previously described methods using a battery of sources of BMZ antigens . IB disclosed weak positive reactivity with recombinant protein (RP) of the BP180 NC16a domain, while IB studies of the BP180 C‐terminal domain RP, concentrated culture supernatant of HaCaT cells, normal human dermal extracts, and purified human laminin‐332 showed negative results.…”

“…Autoimmune bullous disease (AIBD) is a tissue‐specific autoimmune disorder of the skin, and is now classified into many different subtypes . Most of the previous basic studies of various AIBDs were performed to characterize autoantibodies and antigen molecules, and to examine the pathogenic role of autoantibodies using various disease models . In addition, pathomechanisms for the development of autoimmunity in AIBDs have mainly focused on determining the role of T cells and the interaction between T cells and B cells .…”
