Superoxide radical generation and histopathological changes in hippocampal CA1 after ischaemia/ reperfusion in gerbils

Abstract: Toshimitsu Kitajima MDPurpose: We investigated the relationship between the generation of superoxide radicals and histopathological changes on delayed neuronal death in the hippocampal CA I subfield. Methods: Seventy gerbils were randomly assigned to two groups, a sham group and an ischaemia/reperfusion (I/R) group. In the I/R group, transient forebrain ischaemia was induced by occluding the bilateral common carotid arteries for four minutes. The cerebrum was removed after reperfusion at intervals of one minut… Show more

“…Moreover, experimental evidence exists that preinsult hyperglycemia causes an increase in brain damage resulting from transient ischemia (54). A direct correlation between superoxide radical generation and histopathological changes of pyramidal cells after cerebral IR has been reported (55), as has the exacerbation of neurological and histopathological outcomes after focal cerebral ischemia in Cu-Zn superoxide dismutase gene knockout mutant mice (56). Besides the other well-known mechanisms that increase free radical production in diabetes (57), the sharp drop in intracellular pH induced by hyperglycemic ischemia favors free radical production (58).…”

“…Animals were subjected to both common carotid arteries occlusion for 5 min. After the end of ischemia and at different reperfusion times (2,6,12,24,48, 72, 96 hr and 7 days), markers of lipid peroxidation, reduced and oxidized glutathione levels, glutathione peroxidase, glutathione reductase, manganese-dependent superoxide dismutase (MnSOD) and copper/zinc containing SOD (Cu/ZnSOD) activities were measured in hippocampus, cortex and striatum. Oxidative damage in hippocampus was maximal at late stages after ischemia (48 to 96 h) coincident with a significant impairment in glutathione homeostasis.…”

“…Evidence for significant increases in O 2 . has been found as late as 3-4 d after global ischemia in the gerbil (Yamaguchi et al, 1998). The significance of increased O 2 .…”
