Suppression of Intimal Hyperplasia by a 5-lipoxygenase Inhibitor, MK-886: Studies with a Photochemical Model of Endothelial Injury

Abstract: SummaryIntimal thickening is a major complication following percutaneous transluminal coronary angioplasty, which leads to restenosis and requires reoperation. We have investigated the effect of a 5-lipoxygenase inhibitor, MK-886, a leukotriene B4 (LTB4) receptor antagonist, ONO-4057 or a LTC4 and LTD4 receptor antagonist, ONO-1078, on intimal thickening. Photochemical reaction between green light and systemically administered Rose Bengal produced intimal thickening in the rat femoral artery. Each drug was adm… Show more

“…To investigate the impact of MK-886 on I/R-triggered cardiac injury, we first pretreated WT mice with MK-886 at a dosage of 10 or 20 mg/kg and subjected them to I/R as described in previous studies ( Fig. 1 A–B) [ 15 , 16 ]. At 24 h after treatment, measurement of LDH activity (a marker of cardiac injury) indicated that neither dose had a significant cardiotoxic effect in sham mice, whereas 20 mg/kg MK-886 significantly reduced LDH activity in the hearts of I/R mice ( Fig.…”
