Survivin co-ordinates formation of follicular T-cells acting in synergy with Bcl-6

Abstract: Follicular T helper (Tfh) cells are recognized by the expression of CXCR5 and the transcriptional regulator Bcl-6. Tfh cells control B cell maturation and antibody production, and if deregulated, may lead to autoimmunity. Here, we study the role of the proto-oncogene survivin in the formation of Tfh cells. We show that blood Tfh cells of patients with the autoimmune condition rheumatoid arthritis, have intracellular expression of survivin. Survivin was co-localized with Bcl-6 in the nuclei of CXCR5+CD4 lymphoc… Show more

“…Evaluation of clinical symptoms in relation to serological parameters presented a diverse picture, in which survivin was identified adherent to most of them and served a common link to the symptoms associated with both CRP and autoantibodies. This provides further support to the hypothesis that survivin is critical for biological processes foregoing onset of RA ( 40 , 50 , 52 ). Translating those experimental findings into clinical practice, the adherence of early joint symptoms to survivin shows an independent added value of survivin measurement to identify arthralgia patients, that eventually develop RA.…”

“…Mice were immunized with mBSA (Sigma), CII, or OVA as previously described (60,61). Briefly, the antigens were emulsified in complete Freund's adjuvant (Sigma-Aldrich) s.c. on day 0 (200 μg per mouse) followed by a booster emulsified in incomplete Freund's adjuvant on day 7 (100 μg per mouse).…”

“…Samples (1 × 10 5 cells) were acquired on an imaging flow cytometer (ImageStreamX MkII, Amnis) and analyzed using IDEAS version 6.1 software (Amnis) for coexpression of GL7, Flt3, and Bcl-6 as described (61).…”

“…Besides its critical role as a driver of almost every cancer type in humans, increasing data suggest a vital function of BIRC5 for regulating viability and proliferation of non-malignant, physiological adult T cells, which are among the few adult human cell populations with detectable levels of BIRC5 expression (Andersson et al, 2015; Croft et al, 2009; Sharief and Semra, 2001). Within antigen-specific T cells, BIRC5 expression is controlled by OX40 (TNFRSF4, CD134), a member of the TNF receptor superfamily that is expressed on the cell surface and seems to have unique functions for protecting long-term cell viability of memory T cells, particularly at the time of clonal expansion (Song et al, 2005).…”
