Syndecan-1 - A new piece in B-cell puzzle

Abstract: Syndecans are transmembrane proteoglycans, with core proteins mainly decorated with heparan sulfate chains. Syndecan-1 is expressed in a tissue-, cell-and differentiation-specific manner. Its extra-cellular domain can bind via HS chains to matrix elements, to growth factors (especially "heparin-binding" proteins) and to certain biological agents. The ectodomain released by proteolysis can also be functionally active. The cytoplasmic domain can take part in signaling processes as well as in modifying cell shape… Show more

“…We show that normal primary B lymphocytes express a broad repertoire of PG core proteins, with CD44 and serglycin as major PGs. Our data are in line with previously published results showing that CD44 [ 32 ], serglycin [ 9 11 ] and syndecan-1/CD138 [ 15 , 16 ] are expressed in B lymphocytes. In addition we demonstrate for the first time that B cells express additional proteoglycans (perlecan, glypican-1, CSPG4/NG2, decorin, biglycan, lumican) and a wide variety of PGs is consistently altered in EBV-carrying LCLs compared to primary B lymphocytes.…”

“…Thus, syndecan mediates B cell stage-specific adhesion [ 12 , 13 ]. Syndecan is expressed in chronic lymphocytic leukaemia B-CLL, both in tissue environment and in circulation [ 14 , 15 ]. Syndecan expression is not detected in normal and malignant T cells [ 16 ].…”

“…TIN with IgM-positive cells is a new histologic entity with clinical features similar to the present case, such as RTA, Fanconi syndrome, high serum IgM levels and AMA positivity and one of its features is IgM-positive plasma cell (CD138-positive cell) infiltration (13)(14)(15). CD138 is an excellent marker of plasma cells (16,17), but it is also expressed in some B-chronic lymphocytic leukemia (18) and in body cavity-based lymphomas (primary effusion lymphomas) (19). Our case did not show clinical aspects of either of these entities.…”

“…In lymphoid cells, syndecan-1 is expressed in the late stages of differentiation, in pre-B cells and in Ig-producing plasma cells [15,16]. However, the exact role of syndecan-1 on lymphoid cells and the regulation of its expression is still mostly unknown.…”
