Synthesis of leukotrienes in frog retina and retinal pigment epithelium

Abstract: Leukotrienes (LT) were identified in the intact frog (Rana pipiens) retinal pigment epithelium (RPE), retina, isolated rod outer segments (ROS), and ROS-free (neuronal) retina. Levels of endogenous LTC4 were measured by radioimmunoassay (RIA) in both unpurified incubation medium and in incubation medium purified by reverse-phase high-performance liquid chromatography (HPLC). The purified LTC4 exhibited characteristic ultraviolet absorption spectrum with lambda max at 280 nm. The Ca2+ ionophore A23187 (10 micro… Show more

“…Production of cysLTs in whole‐retina preparations appears to be species specific. For example, frog retinas can produce LTC 4 following Ca 2+ ‐ionophore stimulation (44), and rat retinas produce LTB 4 in response to light and trauma (45), whereas cultured bovine pericytes are able to produce cysLTs only in the presence of polymorphonuclear cells, as they produce and donate the LTA 4 (17). Isolated adult mouse retinas require the addition of LTA 4 in order to synthesize LTB 4 , as the tissue in those studies was apparently devoid of 5‐LOX mRNA (15).…”

“…Thus, a very specific free DHA pool may be the precursor for NPD1. Arachidonic acid is also an active precursor of several bioactive lipids, including prostaglandins and lipoxygenase products, and they have been correlated with photoreceptor phagocytosis (31,32). Because arachidonic acid is released under the present experimental conditions (data not shown), it was therefore of interest to explore some of the arachidonic acid cascade members.…”

“…AA is also a precursor of bioactive lipids, including prostaglandins and lipoxygenase products, which have been correlated with photoreceptor phagocytosis (46, 47). That AA is released under the present experimental conditions (data not shown), led us to explore some of the AA cascade members.…”
