Synthesis of β-cyclodextrin conjugated superparamagnetic iron oxide nanoparticles for selective binding and detection of cholesterol crystals

Abstract: Water-soluble, β-cyclodextrin conjugated superparamagnetic nanoparticles have been constructed. These particles showed selective binding to cholesterol crystals, which opens the door for the detection of cholesterol crystal-related diseases such as atherosclerosis by magnetic resonance imaging (MRI).

“…In vitro in HeLa cells, β-CD/chitosan complexes exhibited an enhanced capacity to efflux cholesterol from cell membranes as compared to β-CD alone and the addition of CEEP to the β-cyclodextrin/chitosan complex double this efficacy of cholesterol extraction from cell membranes [52]. In 2012, Li and colleagues [51] conjugated carboxy-methyl-β-CD with superparamagnetic ion oxyde nanoparticles exhibiting a selective binding to CCs with the aim to further use as a detection tool in MRI for atherosclerosis.…”

“…This will be more extensively discussed later in this review for atherosclerosis and neurodegenerative diseases such as Alzheimer's and Parkinson's diseases (Figure 2). In addition, CDs can be used as a carrier enabling the selective binding to biomolecules of interest, as reported for example for cholesterol crystal detection in atherosclerosis ( [51], later discussed in this review).…”

“…Thus, a biological solvent that can accelerate the dissolving of CCs in conjunction with lowering the blood cholesterol levels may represent an opportunity to improve cerebrovascular health in this at-risk population. Recent discovery of β-cyclodextrin conjugated superparamagnetic nanoparticles [37] that selectively bind to CCs will be a vital diagnostic tool for imaging of CCs in CC-related neurological diseases such as atherosclerosis. Importantly, our observation of CC formation in animal model of atherosclerosis was validated in the alcohol environment in human subjects, which has a significant clinical relevance to the health of chronic alcohol consumers.…”

“…Multiple epitopes of the same ligand exposed and a presentation in a three-dimensional format may result in enhanced affinities of the polysaccharide for its biological target 7. As an example, Li and colleagues designed magnetic nanoparticles with multiple copies of cyclodextrin functionalizing the surface in order to enable simultaneous interaction with many cholesterol molecules, leading to high affinity binding with cholesterol for MRI detection purposes 50. In fact, nanosystems may act as platform amplifying the weak affinities of polysaccharide ligands with biological molecules of interest.…”
