Synthetic Archaeosome Vaccines Containing Triglycosylarchaeols Can Provide Additive and Long-Lasting Immune Responses That Are Enhanced by Archaetidylserine

Abstract: The relation between archaeal lipid structures and their activity as adjuvants may be defined and explored by synthesizing novel head groups covalently linked to archaeol (2,3-diphytanyl-sn-glycerol). Saturated archaeol, that is suitably stable as a precursor for chemical synthesis, was obtained in high yield from Halobacterium salinarum. Archaeosomes consisting of the various combinations of synthesized lipids, with antigen entrapped, were used to immunize mice and subsequently determine CD8+ and CD4+-T cell … Show more

“…The resulting lipid was sulfated lactosyl archaeol (SLA; 6'-sulfate-β-D-Galp-(1,4)-β-D-Glcp-(1,1)-archaeol) and was synthesized as described previously [22,23]. M. smithii (MS) total polar lipids were extracted as described previously [22,24,25]. SLA archaeosomes were formed and simply admixed with OVA, hereafter referred to as SLA-OVA (adm) as described previously [26].…”

“…In particular, archaeosomes are thermally resistant and can maintain entrapment integrity even when autoclaved 9 . Archaeosomes are also more susceptible to uptake by phagocytic cells than liposomes of ester phospholipids 10 leading to their utility as adjuvants in the development of antibodies 2,11,12 . The wider use of archaeosomes is directly limited by availability of archaeal lipids derived as mixtures from natural sources 7 .…”

“…Interestingly, a novel SLA archaeosome formulation, whereby the antigen is simply admixed with preformed SLA archaeosomes, has also been shown to stimulate equal or superior antigen-specific responses as formulations where the antigen is entrapped within the archaeosome [20]. As the efficiency of antigen entrapment within archaeosomes formulations is typically variable and relatively low (5-40%) [21,22], this new admixed formulation provides a convenient easy to mix format with no loss of antigen during the formulation process, thereby reducing costs and standardizing the amount of archaeal lipid in the final vaccine formulations.…”
