Systems Vaccinology Identifies an Early Innate Immune Signature as a Correlate of Antibody Responses to the Ebola Vaccine rVSV-ZEBOV

Abstract: SummaryPredicting vaccine efficacy remains a challenge. We used a systems vaccinology approach to identify early innate immune correlates of antibody induction in humans receiving the Ebola vaccine rVSV-ZEBOV. Blood samples from days 0, 1, 3, 7, and 14 were analyzed for changes in cytokine levels, innate immune cell subsets, and gene expression. Integrative statistical analyses with cross-validation identified a signature of 5 early innate markers correlating with antibody titers on day 28 and beyond. Among th… Show more

“…In addition, a systems vaccinology approach identified molecular and cellular immune signatures of a vaccine against Bordetella pertussis [107]. IP-10 was identified as an early innate immune signature that correlated with antibody responses to an Ebola vaccine (rVSV-ZEBOV) [108].…”

“…We investigated innate immune responses following VSV-EBOV immunization and our data revealed an innate immune signature that correlated with antibody responses in the Hamburg VSV-EBOV vaccine trial. 71 In addition, changes in expression levels of circulating miRNAs have been associated with increased EBOV GP-specific antibody titers. Furthermore, innate immune signatures were assessed in vaccinees from Europe and Africa, revealing an early induction at day 1 p.v.…”

“…Development of antigen-specific T cells and their cytokine profiles, as well as the induction of innate immune responses, will need to be directly analysed to understand earlier immune responses. 26,27 Additionally, the correlates of immunity that reflect long-term clinical protection, and whether they are similar or different from those mediating immediate protection, need to be understood. 28 Data from non-human primate models and clinical phase 1 studies of combined Ad26.ZEBOV and MVA-BN-Filo vaccination 15 provide evidence of an important role for cellular immunity, particularly CD8-positive T cells producing tumour necrosis factor α and interferon γ (with or without interleukin 2).…”
