T-cell and antibody immunity after COVID-19 mRNA vaccines in healthy and immunocompromised subjects-An exploratory study

Abstract: Background: In recent studies, up to half of immunocompromised (IC) subject populations fail to develop antibodies after COVID-19 vaccination. Purpose and Methods: Here, we explore whether T-cells which respond to the spike (S) antigenic sequence and its less conserved S1, and the conserved S2 component are present in serial samples before and after each dose of mRNA1273 or BNT162b2 vaccines in 20 healthy immunocompetent subjects. Single samples from 7 vaccinated IC subjects were also tested. Simultaneously, w… Show more

“…The T-cell response rate in our population was extremely low, also when compared to a previous study [12]. This could be caused by the assay's performance, or the fact that SOT recipients are heavily treated with Tcell immunosuppressants-especially calcineurin inhibitors [13]. The true T-cell response might be underestimated, and our results need to be further studied in a larger cohort.…”

“…All 19 studies described participant age and sex. Five studies (26%) described the distribution of participant race or ethnicity ( 21 , 23 , 24 , 25 , 26 ), and 4 studies (21%) described multimorbidity ( 20 , 27 , 28 , 29 ). Two studies (11%) focused on assessing response to vaccination in health care workers; thus occupation was described ( 20 , 27 ).…”

“…The S antigenic sequence is made up of the less conserved N-terminal S1 component, which contains the receptor-binding domain (RBD) and the conserved C-terminal S2 sequence. 15 It binds to the host cell’s ACE2 receptor and causes membrane fusion. 10 Antibodies that bind to the RBD of the SARS-CoV-2 spike protein can prevent the virus from attaching to the host cell and neutralize it.…”
