T cell proliferation in the mixed lymphocyte culture does not necessarily result in the generation of cytotoxic T effector cells

Abstract: It was tested whether T lymphocytes, when stimulated in vitro by M locus-coded lymphocyte activating determinants (LAD), are able to mediate cytotoxic effector functions. The assay for cytotoxicity included both the use of purified appropriate target cells (i.e. purified lipopolysaccharide blasts) as well as the use of phytohemagglutinin dependent cytolysis as a model for detecting cytotoxic T lymphocytes (CTL). Although strong proliferative responses were obtained in the mixed lymphocyte culture, the T cell b… Show more

“…Mls-la is expressed primarily on B cells [12,131 and stimulates mainly Vp6+ and to a lesser extent I439+, V&l+ and I437+ cells [14-181. Responses to Mls-1" were originally considered to be restricted to CD4+ cells, since they were inhibited by CD4 specific antibodies and dependent on presentation by MHC class I1 molecules [19]. Consistent with this assumption was the failure to detect Mls-la-specific cytotoxicity but, retrospectively, this could have been due to the choice of putatively CD4+ cells as effectors [20] [25]. Furthermore, this recognition was not inhibited by CD8-specific mAb 12.51.…”

“…Mice. C3H and C57B1/6 from Bomholtgaard, Ry, Denmark, were used at an age of [8][9][10][11][12] weeks. The Ly congenic mouse strains C57B1/6-Ly-ia/Boy, abbreviated B6/Ly-l.l; C57Bl/6-Ly-2a/Boy, abbreviated B6/Ly-2.1; and C57Bl/6-Ly-2aLy-3a/Boy, abbreviated B6/Ly-2.1, Ly-3,1 were originally kindly provided by Dr. E. A. Boyse (Memorial Sloan Kettering Cancer Center, New York, N.Y,) and further propagated by strict brother-sister mating in our own mouse facilities.…”
