Targeted Delivery of Cabazitaxel Using Cyclic Cell-Penetrating Peptide and Biomarkers of Extracellular Matrix for Prostate and Breast Cancer Therapy

Abstract: Targeted drug delivery for cancer therapy is an emerging area of research. Cancer cells overexpress certain biomarkers that can be exploited for their targeted therapy. Cyclic cell-penetrating peptides (cCPP) are increasingly assessed for intracellular cargo delivery in cancer cells. In this study, we have conjugated cabazitaxel (CBT) to the cCPP via an ester bond to assist CBT release in the tumor's acidic environment. Integrin targeting (RGDC, TP1) and extra domain B of fibronectin (EDB-Fn) targeting (CTVRTS… Show more

“…Active targeting of small molecule drugs is essential to improving the efficacy of antitumor therapy. Park et al ( 2021 ) synthesized cyclic CPP conjugated CBT (cCPP-CBT) via an ester bond that can be hydrolyzed by esterase. They also conjugated targeting peptides with cCPP-CBT (TP-cCPP-CBT) via a disulfide bond that can be labile under a reductive environment.…”

“…SP90-C revealed well targeted delivery function for anticancer drugs. Park et al (2021) synthesized anticancer drugs targeting the weak acidic condition of the breast tumor by conjugating cyclic CPP (cCPP, CWRWRKWRWR) with a targeting peptide (TP1 or TP2) and anticancer drug cabazitaxel (CBT). TP1-cCPP-CBT revealed antiproliferative activity on MDA-MB-231 and MCF-7 cells.…”

“…The conjugates showed less toxicity to normal human embryonic kidney (HEK-293) cells compared to free CBT while displaying approximately three- to four-fold higher antiproliferative activity on cancer cell lines, compared to the free CBT analog. Although the increased drug delivery can be attributed to the presence of CPPs, the widespread use of these types of PDCs is still limited due to their low cellular specificity [ 91 ].…”
